Effects of Spent Mushroom Compost (SMC) as an Ingredient in Phase I Compost on Production of Agaricus bisporus

ABSTRACT

Experiments were performed to determine the effect of using 20% spent mushroom compost (SMC) as an ingredient in phase I compost on mushroom yield. Yields from compost prepared with two 20% SMC formulations (80Ctl, 80LC) were compared to a standard control (Ctl) containing no SMC. The 80LC compost was formulated with a higher level of lignocellulose raw materials, including corn stover, corncobs, cottonseed hulls, and wheat straw. The 80Ctl compost contained 20% SMC plus the same raw materials used to prepare Ctl compost. Yields of 80LC matched or exceeded yields of the Ctl compost in all three crops while yields of 80Ctl were significantly lower. As an additional factor, lignocellulose supplements were added at spawning and were a significant factor in increasing mushroom yields, although not superior to a standard commercial supplement applied at 3.7% (dry compost weight). Compost bulk density was significantly higher (19–20%) in compost formulations containing 20% SMC compared to the non-SMC Ctl. Populations of four prominent phyla of bacteria (Firmicutes, Actinobacteria, Proteobacteria, and Chloroflexi), as determined by pyrosequence analysis, were similar in both compost types and did not appear to influence mushroom yield.

Comparison of characterization and microbial communities in rice straw- and wheat straw-based compost for Agaricus bisporus production

Abstract

 

Rice straw (RS) is an important raw material for the preparation of Agaricus bisporus compost in China. In this study, the characterization of composting process from RS and wheat straw (WS) was compared for mushroom production. The results showed that the temperature in RS compost increased rapidly compared with WS compost, and the carbon (C)/nitrogen (N) ratio decreased quickly. The microbial changes during the Phase I and Phase II composting process were monitored using denaturing gradient gel electrophoresis (DGGE) and phospholipid fatty acid (PLFA) analysis. Bacteria were the dominant species during the process of composting and the bacterial community structure dramatically changed during heap composting according to the DGGE results. The bacterial community diversity of RS compost was abundant compared with WS compost at stages 4–5, but no distinct difference was observed after the controlled tunnel Phase II process. The total amount of PLFAs of RS compost, as an indicator of microbial biomass, was higher than that of WS. Clustering by DGGE and principal component analysis of the PLFA compositions revealed that there were differences in both the microbial population and community structure between RS- and WS-based composts. Our data indicated that composting of RS resulted in improved degradation and assimilation of breakdown products by A. bisporus, and suggested that the RS compost was effective for sustaining A. bisporus mushroom growth as well as conventional WS compost.

Effect of compost compression, perforation in compost bags and watering schedule on yield of white button mushroom, Agaricus bisporus

Abstract

The study presents the results of compost compression, perforation in compost bags and the watering regime on the fruit body yield of the white button mushroom, A. bisporus. In four cultivation trials performed during different periods of three years in 10 kg compost bags, least time for first harvest (days post-casing) was recorded in compost compressed up to 3" along with perforation in compost filled polybags @ 30 holes of 0.5 cm dia / bag. In different compost compression treatments, the first harvest was earlier and also fruit body yield (kg/ 100kg compost) was higher in bags with perforation compared to bags without perforation. The yield was highest in bags without any compression of compost but with perforation, followed by bags compressed up to 5" depth along with perforation. The average fruit body wt. was above 13 g in all the treatments but in most of the cases it was higher in bags with perforation in compost bags. The spent mushroom substrate from the high yielding treatments was found to have higher nitrogen content than in treatments with lower yield. Heavy watering on day 0, 4th and 7th, followed by routine spray of water gave higher fruit body yield.

Biochemical properties of different agricultural wastes treated with Pleurotus ostreatus

Abstract

An analytical study of  different agricultural wastes namely, wheat straw, paddy straw, cotton waste, sugarcane baggasse and banana leaves was performed which included analysis of crude protein (%), crude nitrogen (%), crude fiber (%) and ash contents. These contents were estimated in all the substrates, to make the comparisons that how the cultivation of Pleurotus ostreatus led to enhancement of its contents. The mean values of the contents were noted before and after inoculation with the Pleurotus ostreatus spawn. The mean values of the ash content were found to be the maximum in cotton waste and least in banana leaves. Similarly in case of nitrogen, the mean values is found maximum in cotton waste followed by wheat straw. The highest protein content (%) was found in cotton waste followed by wheat straw, banana leaves, paddy straw and least content was in sugarcane baggasse. The mean values for the crude fiber is found maximum in cotton waste followed by sugarcane baggase, wheat straw, paddy straw and the least amount is in banana leaves. The analysis of the substrate showed the enhancement of their nutritional values.

ABSTRACT

Supplementation of substrate at casing to increase the yield and quality
of mushroom [/Agaricus bisporus/(Lange) Sing] is an important practice
in commercial production of white button mushroom. This project was done
to study the effects of supplementing the compost at casing with ground
corn and soybean seed applied at: 0 gas control, 17, 34 and51 gper17
kgcompost on production and harvest quality of /A//. bisporus/. There
were significant differencesbetween supplemented and non-supplemented
substrates. The34 gsoybean and51 gcorn treatments had the highest yield.
There were significant differences in quality indices of mushroom due to
the type and amount of the supplement. The51 gsoybean supplement
produced higher protein compared with other substrates. The highest
vitamin C, total phenol, total soluble solids and antioxidant capacity
obtained of 34 and51 gsoybeans or 34 and51 gcorn, respectively.
Increased production and quality of /A//. bisporus/may be achieved by
addition of suitable amounts of supplements

    Abstract

Dry bubble (caused by /Lecanicillium fungicola/) is a widespread disease
of button mushroom. The objective of the experiments was to determine
the efficacy of compost teas made from spent mushroom substrate (SMS) as
a biocontrol method against the disease. All SMS teas produced in this
study significantly inhibited (100 %) the in vitro mycelial growth of
/L. fungicola/, whereas the fungicide prochloraz at 50 ppm inhibited
growth by 91 %. The in vivo effectiveness of two SMS aerated teas, one
with mineral soil (MS) and the other with peat (TPT), was evaluated in
two mushroom cropping trials inoculated with /L. fungicola/. The results
demonstrated that the most effective treatments were those with TPT
applied close to harvest and/or those with the greatest number of
applications. The most efficacious treatments were TPT treatments
(reducing disease by 34 to 73 % in the two trials, compared to the
inoculated control). In contrast, prochloraz reduced disease by 7 % and
4 % in the two trials, compared to the control. These results suggest
that dry bubble disease can be controlled by the use of spent mushroom
substrate teas.

    Abstract

Casing materials and practices used in the cultivation of /Agaricus
bisporus/ were evaluated in the cultivation of /Agaricus subrufescens/,
using the best techniques for optimization of production, including the
possibility of re-casing of the compost for the production of a second
crop of mushroom. Casing based on peat moss, loam soil or coir was
compared to casing material mixed with or without spawn-run compost.
Based on the results, we conclude that the casing layer used in the
cultivation of /A. subrufescens/ should not necessarily be the same as
that used in the cultivation of /A. bisporus/. For the tested strain
cultivated with loam soil as casing layer, the ruffling technique is
highly superior to CACing and should be pursued in further research. The
re-casing of compost in new cycles showed good results suggesting that
the currently used compost could be improved.

        شناسایی اکتینومیست‌های کمپوست قارچ خوراکی دکمه‌ای - خاک پوششی
        کمپوست قارچ

آکتینومیست‌ها، بویژه گونه‌های گرمادوست، بعنوان اصلی‌ترین اجزای میکروفلور
کمپوست قارچ خوراکی تکمه‌ای شناخته‌شده‌اند. آکتینومیست‌های گرمادوست با
استفاده از روش رقیق‌سازی بر روی محیط کشت آگار و انکوبه شدن در دمایC 48
جداسازی شدند. مقایسه الگوی قطعات برشی ایزوله‌های جداشده با الگوی تهیه
شده از هضم تمامی توالی‌های معتبر منتشر شده از گونه‌های آکتینومیست‌ها به
صورت این سیلیکو، روشی سریع برای شناسایی آکتینومیست‌ها می‌باشد. جهت تفکیک
ایزوله ها، جزء کوچک ژن RNA ریبوزومی (16SrRNA) به طول تقریبی 1500 جفت باز
از آکتینومیست‌های جداسازی شده با استفاده از واکنش زنجیره‌ای پلیمراز و
پرایمر‌های عمومی fD1 و rP2 تکثیر شد. با استفاده از آنزیم‌های برشی، ژن
16SrRNA تکثیر شده هر ایزوله، جداگانه هضم و الگوی برشی هر آنزیم با
الکتروفورز روی ژل آگارز تعیین گردید. از تکنیک تجزیه و تحلیل قطعات DNA
ریبوزومی تکثیر شده ( ARDRA) برای شناسایی ایزوله‌های ناشناخته استفاده شد.
بیشترین آکتینومیست‌های جداسازی شده از کمپوست قارچ خوراکی تکمه‌ای به جنس
استرپتومایسس تعلق داشتند که از اهمیت زیادی در تجزیه بقایای گیاهی و تولید
کمپوست برخوردارند

  Anaerobically digested food waste in compost for /Agaricus bisporus/
  and /Agaricus subrufescens/ and its effect on mushroom productivity
 


      Abstract


        Background

*Source-separated food waste is increasingly being treated by means of
hygienisation followed by anaerobic digestion. The fibrous digester
residue (digestate) is a potential mushroom substrate, while heat from
the biogas can provide steam for the cultivation process. Using bag
experiments the present study explored digestate as a full substitute
for chicken manure conventionally used in mushroom composts*.


        Results

*After mixing, a rapid temperature development in the compost was
stimulated by a small amount of chicken manure, as aerobic microbial
seeding. Mechanical elimination of lumps was essential for full mycelial
colonisation. Three straw digestate composts had /Agaricus bisporus/
mushroom yields above 370 g kg^−1 substrate. The optimal compost water
content was 600 g kg^−1 at inoculation, and high digestate content (up
to 500 g kg^−1 by dry weight) did not affect yield for this species.
High yields of /A. subrufescens/ (200 g kg^−1 ) were related to drier
composts of lower digestate content (more straw) and lower pH values at
inoculation*.


        Conclusion

*Digestate successfully substituted chicken manure in straw composts
without affecting mushroom yields for both species. There were no clear
differences between straw digestate and control composts in terms of
mushroom dry matter, size, nitrogen or ash content. © 2013 Society of

      Abstract

The effect of the agent of dry bubble disease /Lecanicillium fungicola/
on mushroom cultivation using two types of casing (mineral soil and
peat-based) was evaluated in two artificially infected cropping trials.
The incidence of dry bubble and loss of yield caused by the disease were
assessed. Biological efficiency (BE) was used to evaluate mushroom
productivity for the different casing soils. The results showed that, in
the absence of /L. fungicola/ infection, the higher BE obtained using
peat-based casing (higher yield of /A. bisporus/) compensates for the
higher cost of the casing. However, when facing serious infection by /L.
fungicola/, the incidence of disease and the yield of healthy mushrooms
were statistically similar for both types of casing, although the number
of diseased mushrooms was higher when peat-based casing was used.
Therefore, the use of peat-based casings cannot be recommended when
there is a high risk of outbreaks of dry bubble, since the yield of
healthy mushrooms is not sufficient to compensate for the difference in
price between both casing types.

  Inactivation of Human Pathogens during Phase II Composting of
  Manure-Based Mushroom Growth Substrate
 

*Abstract:*

Commercial production of white button mushrooms (/Agaricus bisporus/)
requires a specialized growth substrate prepared from composted
agricultural by-products. Because horse and poultry manures are widely
used in substrate formulations, there is a need to determine the extent
to which the composting process is capable of eliminating human
pathogens. In this study, partially composted substrate was inoculated
with a pathogen cocktail (log 10^6 to 10^8 CFU/g) containing /Listeria
monocytogenes, Escherichia coli/ O157:H7, and /Salmonella/. Pathogen and
indicator-organism reductions were followed at temperatures that
typically occurred during a standard 6-day phase II pasteurization and
conditioning procedure. Controlled-temperature water bath studies at
48.8, 54.4, and 60°C demonstrated complete destruction of the three
pathogens after 36.0, 8.0, and 0.5 h, respectively. Destruction of /L.
monocytogenes/ and /E. coli/ O157:H7 at 54.4°C occurred more slowly than
/E. coli,/ total coliforms, /Enterobacteriaceae,/ and /Salmonella/.
Microbial reductions that occurred during a standard 6-day phase II
pasteurization and conditioning treatment were studied in a small-scale
mushroom production research facility. After phase II composting, /E.
coli,/ coliforms, and /Enterobacteriaceae/ were below detectable levels,
and inoculated pathogens were not detected by direct plating or by
enrichment. The results of this study show that a phase II composting
process can be an effective control measure for eliminating risks
associated with the use of composted animal manures during mushroom
production. Growers are encouraged to validate and verify their own
composting processes through periodic microbial testing for pathogens
and to conduct studies to assure uniform distribution of substrate
temperatures during phase II.

  The effect of growth promotion and its mechanism on edible fungi by
  fluorescent Pseudomonas

fluorescent pseudomonads can promote mycelia growth and development of
some mushroom, such as Agaricus bisporus, Pleurotus ostreatus, and
Pleurotus eryngii. It remains poorly characterized whether fluorescent
pseudomonas has similar effect on other cultivated mushroom and what is
the mechanism. A strain of fluorescent pseudomonad was isolated from
wheat rhizosphere and named Pseudomonas sp. P2-10. The level of partial
16S rDNA sequence similarity between strain P2-10 and the fluorescent
Pseudomonas species from GenBank ranged from 94% to 98%. The highest 16S
rDNA sequence similarity (98%) was observed between strain P2-10 and P.
putida IAM 1236T, P. Koreensis Ps 9-14T, P. pavonaceae IAM 1155T, P.
jessenii CIP 105274T, and P. umsongensis Ps 3-10T. With addition of
bacterial culture strain P2-10 and its supernatant to cottonseed hull
media, mycelia of P. ostreatus Td 300, P. eryngii X6, and Coprinus
comatus R7 grew faster and fruit body yield of P. ostreatus and P.
eryngii were significantly higher than that of control. Similarly adding
P2-10 culture to axenic casing soil of C. comatus R7 also stimulated
basidiome initiation. The substance for those effects was in cultural
solution of strain P2-10, and was similar to that of ethylene inhibitor
AgNO3 and CoCl2. Strain P2-10 contained the enzyme
1-aminocyclopropane-1-carboxylate (ACC) deaminase and can utilize ACC as
a sole source of nitrogen. Results of this research strongly suggest
that inoculation of the mycelia with the fluorescent pseudomonas may
promote mycelia growth, basidiome initiation, and enhance fruit body
yield of P. ostreatus, P. eryngii, and C. comatus. The mechanism may be
that ACC deaminase degrades ACC and decreases ethylene synthesis by
edible fungi, subsequently relieves the inhibitory action of ethylene to
mycelia growth and fruit body development of edible fungi.

* An improved rapid composting procedure enhance the substrate quality
and yield of Agaricus bisporus*


 

The effect of passive aeration on substrate quality, composting period
and yield was studied using different perforated high-density
polyethylene (HDPE) pipes arrangements, as a composting method for
substrate preparation to Agaricus bisporus cultivation. The ingredients
are, mixture of wheat straw (60%), wheat bran (5.5%), chicken manure
(31%), urea (0.5%) and gypsum (3%) with initial moisture (75%) used for
composting. It was demonstrated that passive aeration is the key
parameter controlling composting process, shortened the compost period,
enhance the substrate quality and Agaricus yield as of pile composting.
The substrate physico-chemical characteristics that is, moisture
content, temperature pattern, bulk density, pH, electric conductivity,
C: N ratio and thermophilic fungi activity varied significantly with
composting period. The parallel arrangement (10% perforations) was found
best among all the passive aeration treatments for achieving the quality
substrate in shortest composting period of 16 day and enhance the A.
bisporus yield up to 27.6%. This study also opens possibility to
cultivate A. bisporus on a lingo-cellulosic, non-pasteurized,
non-conditioned, aerated substrate and that composting for 16 days
improve the mushroom yield with minimum energy, labor and infrastructure.

  Role of Thermophilic Microflora in Composting
 


    Abstract

Composting is a process that converts organic waste to a humus-like end
product and is not only a waste treatment technique but also a recycling
method as the end product can be used in agriculture as fertiliser, in
gardening or in landscaping. Microbes play a key role as degraders
during the composting process; mesophilic microflora constitutes the
pioneer component, while thermophilic microflora, the climax and also
the dominant component contributes significantly to the quality of
compost. The dynamics of microbial community in different compost
ecosystems change in context with qualitative and quantitative changes
in physico-chemical conditions of compost. Optimisation of compost
quality is directly linked to composition and succession of microbial
communities in compost ecosystem. This necessitates the monitoring and
characterisation of the microbial community composition, patterns and
dynamics of species diversity at spatial scales. The biasedness and
time-consuming nature of cultivation-based approaches severely limits
spatial and temporal intensity of sampling, but with the advent of
culture-independent molecular techniques, new insights into the
composition of uncultivable communities have been gained. It has also
been now possible to define the causes of time-dependent changes in the
health of microbial community on the basis of observed 16S and 18S rDNA
diversity. The genomic information represented by such a ‘community
genome’ offers a tremendous resource for examining the extent and
patterns of microbial genetic diversity and metabolic capabilities in
the natural ecosystem. These studies also provide basic knowledge on
mechanisms that control species coexistence, which have fundamental
applications since they offer the framework that serves in maintaining,
restoring and manipulating the diversity in natural compost ecosystem
and hastening the process of composting besides improving the quality of
compost.
