In-vitro antioxidant activities of an ethanolic extract of the oyster mushroom, Pleurotus ostreatus

Abstract

The antioxidant potential of an ethanolic extract of the oyster mushroom, Pleurotus ostreatus, was investigated. The extract exhibited the most potent radical-scavenging activity at a maximum concentration of 10 mg/ml, and the scavenging effects were 56.20% and 60.02% on hydroxyl and superoxide radicals, respectively. The IC50 values of the extract were found to be 8 mg/ml for hydroxyl and superoxide radicals. Ascorbic acid used as a standard was highly effective in inhibiting hydroxyl and superoxide radicals, showing IC50 values of 6 mg/ml and 4 mg/ml respectively. At a maximum concentration of 10 mg/ml, the extract effected 56.12% inhibition of lipid peroxidation and 60.68% chelation of ferrous ions; also, at a maximum concentration 10 mg/ml, the extract manifested significant (p < 0.05) reducing power (1.367) which exceeded even that of butylated hydroxyl toluene (1.192). Increasing concentrations of the extract were found to cause progressively decreasing intensity of fluorescence 2, 3-diazabicyclo [2, 2, 2] oct-2-ene (DBO). In addition, the known antioxidants were identified as components of the extract. The data generated by this study strongly suggest that an ethanolic extract of the oyster mushroom, P. ostreatus, has potent antioxidant activity.

Industrial relevance

The present study suggests that an ethanolic extract of the mushroom, Pleurotus ostreatus, could serve as an easily accessible item of food rich in natural antioxidants, as a possible food supplement or even as a pharmaceutical agent. Hence this study is considerable relevant to the food and pharmaceutical industries.

Structural characterization and in vitro antitumor activity of a novel polysaccharide isolated from the fruiting bodies of Pleurotus ostreatus

Abstract

A novel water-soluble polysaccharide (POPS-1) was obtained from the fruiting bodies of Pleurotus ostreatus by hot water extraction, ethanol precipitation, and fractionated by DEAE-cellulose ion exchange chromatography and sepharose CL-6B gel filtration chromatography using an ÄTKA explore 100 purifier. The structure characterization and antitumor activity of the POPS-1 were evaluated in this paper. According to GC analysis, HPGPC, FT-IR, partial acid hydrolysis, periodate oxidation and Smith degradation, methylation and GC–MS analysis, the results indicate POPS-1 (Mw = 31 kDa) was composed of Man; Gal; Glc with a molar ratio of 1:2.1:7.9, it had a backbone of β-(1 → 3)-linked glucose residues, which occasionally branches at O-6. The branches were composed of (1 → 3)-linked Glc, (1 → 4)-linked Gal, (1 → 4)-linked Man, and terminated with Glc and Gal residues. Cytotoxicity assay showed POPS-1 presented significantly higher antitumor activity against Hela tumor cell in vitro, in a dose-dependent manner, and exhibited significantly lower cytotoxicity to human embryo kidney 293T cells than Hela tumor cells compared with 5-Fu. The results suggest POPS-1 may be considered as a potential candidate for developing a novel low toxicity antitumor agent.

Screening of Pleurotus Ostreatus and Gleophylum Sepiarium Strains for Extracellular Protease Enzyme Production

Abstract

The fungal isolates identified as Pleurotus ostreatus and Gleophylum sepiarium isolated from sawdust dump-site were screened for protease enzyme production. High yields of protease enzyme were obtained by both fungi after 96h with concentrations of 1.6ug/ml/min for P ostreatus and 1.5ug/ml/min for G.sepiarium. Optimum temperature for the activity of protease produced by P.ostreatus and G.sepiarium was at 70oC with activity of 060ug/ml/min and 0.55ug/ml/min respectively. Optimum pH for the activity of protease produced by P.ostreatus and G.sepiarium was pH 7 with activity of 0.55ug/ml/min and 0.60ug/ml/min. The result showed that both isolates were good producers of extracellular protease enzyme which may be useful in industries.
