Effect of Temperature and Button Mushroom Varieties on Life History of Lycoriella auripila (Diptera: Sciaridae)

Abstract:
Temperature-dependent development and population growth parameters of the fungus gnat, Lycoriella auripila (Winnertz), on button (737 and A15) mushroom were evaluated at nine constant temperatures, ranging from 8, 10, 12.5, 15, 20, 22.5, 25, 27, and 30°C and developmental rates were modeled as a function of temperature. At 25 and 27°C, an average of 20.89 and 22.43 d was needed for L. auripila to complete its development from oviposition to adult eclosion on 737 and A15, respectively. The population failed to survive at 8 and 30°C. The developmental times of males or females on each variety at various constant temperatures differed significantly. The regression and Ikemoto and Takai linear models, in the absence of 10, 12.5, and 27°C, described the relationship of developmental rate to temperature for male and female of L. auripila very well. Data were fitted to various nonlinear temperature-dependent models. The Logan 6 and Briere 1 nonlinear models provided the best estimation for Topt and Tmax and are highly recommended for the description of temperature-dependent development of L. auripila on 737 and A15, respectively. On 737 variety, significant differences were observed among all growth parameters at various temperatures. The intrinsic rate of natural increase (rm) for L. auripila was 0.134 on 737 variety at 25°C, which was significantly >0.052 on A15 variety at the same temperature. No other data are available in using linear and nonlinear models to describe the relationship between temperature and L. auripila development. Understanding the influence of temperature on development of L. auripila is discussed with respect to pest management in mushroom production.

Agaricus Bisporus: Enzymatic browning and its inhibition methods
Browning button mushroom of Agaricus Bisporus is a common phenomenon which enzymaticaly convert melonogenic phenols to quinones and eventually produce melanin pigments. Browning button mushroom is detrimental to business of agriculture and food business. This review article surveys methods of inhibiting the enzymatic browning Agaricus Bisporus include: thermal inactivation, high isostatic pressure, gamma irradiation, thiol componds, calcium chloride added to the irrigation water, inactivators and inhibitors and its effects on enzymatic browning of Agaricus Bisporus. Induction of apoptosis of gastric cancer cells SGC7901 in vitro by a cell-free fraction of Tibetan kefir
Abstract
In this study, the anti-proliferative effect of cell-free fraction of Tibetan kefir on human gastric cancer cell line SGC7901 in vitro was investigated. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed a dose-dependent antiproliferative effect, and when the concentration of Tibetan kefir cell-free fraction was more than 8 mg mL−1, a distinct anti-proliferation of SGC7901 cells was observed. Flow cytometric analysis was carried out to detect cell cycle arrest and apoptosis. SGC7901 cells treated with a cell-free fraction of Tibetan kefir were inhibited in the G1/S phase, and both early and late apoptosis could be detected. To further investigate the mechanism of action of cell-free fraction of Tibetan kefir on apoptosis of SGC7901 cells, expression of human bcl-2 and bax genes were measured by qPCR. Apoptosis induced by Tibetan kefir was effected through up-regulation of bax and down-regulation of bcl-2.

Abstract
The effect of the agent of dry bubble disease Lecanicillium fungicola on mushroom cultivation using two types of casing (mineral soil and peat-based) was evaluated in two artificially infected cropping trials. The incidence of dry bubble and loss of yield caused by the disease were assessed. Biological efficiency (BE) was used to evaluate mushroom productivity for the different casing soils. The results showed that, in the absence of L. fungicola infection, the higher BE obtained using peat-based casing (higher yield of A. bisporus) compensates for the higher cost of the casing. However, when facing serious infection by L. fungicola, the incidence of disease and the yield of healthy mushrooms were statistically similar for both types of casing, although the number of diseased mushrooms was higher when peat-based casing was used. Therefore, the use of peat-based casings cannot be recommended when there is a high risk of outbreaks of dry bubble, since the yield of healthy mushrooms is not sufficient to compensate for the difference in price between both casing types.

Purification and Characterization of Polyphenol Oxidase from Agaricus Bisporus
ABSTRACT
A polyphenol oxidase (PPO) was isolated from Agaricus bisporus using ammonium sulfate precipitation, combined with DEAE–Sepharose Fast Flow chromatography and Phenyl Sepharose CL-4B chromatography. The molecular weight of the purified PPO was determined to be about 43 kDa with N-terminal sequence Ala-Thr-Asn-Ser-Gly-Thr-Leu-Ile-Ile-Phe by Edman degradation. The optimum temperature and pH for the PPO activity was 20 °C and pH 6.5-7.0. Moreover, it was stable at 30 °C and 40 °C over a 60 min pre-incubation time period. The Km and Vmax values of this PPO for catechol were 0.67 mM and 3333 U/ml min−1, respectively. In addition, following the cross linking with PPO, the emulsifying activity index, foam capacity and foam stability of the whey proteins were evaluated to be modified. This work was useful to better understand PPO from A. bisporus, and may lead to its practical use in the future.

Pest Management on Us Commercial Mushroom Farms

Abstract:
In 2011, mushroom production in the United States was valued at approximately $1.10 billion. Total mushroom production was second only to China. Agaricus bisporus production accounts for nearly 98% of the total US mushrooms grown based on volume of sales. Approximately 65% of the US production occurs in Pennsylvania with California producing 14% and the remaining production spread across the US in several states. The close proximity of the Pennsylvania mushroom farms to one another has many advantages for the mushroom growers: centralized composting operations, centralized packing houses as well as having specialized crews that can travel between farms doing a variety of operations that are essential in mushroom production. However, the close proximity of farms, centralized operations and contracted work crews can also lead to many challenges to growers when it comes to disease control. In addition to these challenges, the US mushroom growers have recently been experiencing the loss of many conventional pesticides to address their needs. Fewer chemical options are available today to control pathogens and flies on farms. Because of these challenges, US mushroom growers have been proactive in addressing disease management through an aggressive Integrated Pest Management (IPM) approach. An effective mushroom IPM program utilizes a multiple control approach including exclusion, cultural controls, sanitation, chemical control and biological control measures. The origination of IPM in the US mushroom industry started with a program to address sciarid fly populations in the 1970s. Since then, it has developed into a complete program to address all potential pathogens and pests. To understand the practices utilized by US mushroom growers better, the IPM program will be discussed sequentially, by describing control measures currently being utilized with an explanation of the potential pathogens and pests addressed by each step. The description that follows is not meant to be inclusive of all steps that are implemented on US mushroom farms, but rather it is meant to be a guide to promote the thought process that will provide growers with the tools necessary to improve upon their own IPM program.

Experimental Study of Ozone Treatment System for Mushroom Substrates Sterilization

Abstract:
This paper reports that ozone sterilizing system as the efficient system for spawn treatment of mushroom cultivation. The long time consuming in steam sterilizing system induced the idea to find another efficient sterilize agent for spawn treatment in term of time consuming. This project involves the ozone sterilizing system design, laboratory experiment and analysis of experiment data to validate the use of ozone in mushroom industry. The sterilizing chamber was specially designed in a fashion that mushroom substrate can be injected by ozone simultaneously. It is useful for commercial application for mushroom cultivation. This new system proved that less time will be consumed. Thus, more mushroom substrate can be produced and at the same time the nutrient inside the substrate is increased due to the oxidation process with organic matter. Consequently, more mushroom can be cultivated at a much lower production cost and can be sold at lower price.

Effects of aqueous extracts from Pleurotus sajor-caju on the inhibition growth of breast cancer cell lines

Abstract
Introduction: Cancer is one of the major non-infection health problem affect worldwide including Thailand. Several anticancer drugs have been orginated from natural products. Pleurotus sajor-caju, an edible mushroom, commonly used for sources of nutritional foods in many countries including Thailand, belongs to the Pleurotaceae family and also has commercial cultivation with low cost of production. It has been reported that the Pleurotaceae family have some bio-active effects, such as anti-oxidative, antiatherosclerotic, immunomodulatory and anticancer activities. Few studies have been explored in P. sajor-caju as anticancer pharmaceutical source. In this study, we primarily investigate the effects of an aqueous extracts from P. sajor-caju on the growth of human breast cancer cell line (MCF-7), human liver carcinoma cell lines (HepG2) compared to those of normal cells line (Vero cell). Material and methods: Aqueous extract of P. sajor-caju was lyophilized to dryness. Human breast cancer cell line (MCF-7), human liver carcinoma cell lines (HepG2), and Vero cell (normal cell) were used. The cytotoxic activities of the compounds were assessed using the (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The comparison between cytotoxic effects of the extract and doxorubicin in the corresponding cell lines were also investigated. Results: The results demonstrated that an aqueous extract of P. sajor-caju had cytotoxic effects with 50% cell growth inhibition (IC50) against MCF-7, HepG2 and Vero cell at 200.96±15.42, 438.76±67.12 and 364.96±70.6 µg/mL, respectively, whereas the cytotoxic effects of doxorubicin against MCF-7 and HepG2 showed an IC50 at 3.50±0.60, and 0.99±0.20 µg/mL, respectively. The extract had significantly cytotoxic activity on MCF-7 compared to those of normal cell line at a concentration of 250 and 500 µg/mL (p<0.005). Conclusion: The results demonstrated that an aqueous extract of P. sajor-caju exerts anti-proliferative action on breast cancer cell line MCF-7, suggesting its anticancer properties. Therefore, the further study on the characterization of active compounds and the mechanisms underlying of the anticancer properties induced by P. sajor-caju would be extensively investigated. This finding presented could provide interesting path for further investigation of a novel anticancer agent.
